You can culture it like you would any old staph or strep or what have you, but it divides extremely slowly. Instead it's better to identify it under the microscope with a ZN stain, but again because it replicates so slowly it's often a needle in a haystack situation. With most organisms, you don't have to search for the needle, because you'd see much higher numbers of the organism if it was clinically significant- TB on the other hand is extremely infections, and exposure to literally single digit numbers of cells can cause the disease.
